چکیده

MOTIVATION The concentration of pathogen DNA in biological samples is often very low. Therefore, the sensitivity of diagnostic tests is always a critical factor. RESULTS We have developed a novel computational method that identifies species-specific repeats from microbial organisms and automatically designs species-specific PCR primers for these repeats. We tested the methodology on 30 randomly chosen microbial species and we demonstrate that species-specific repeats longer than 300 bp exist in all these genomes. We also used our methodology to design species-specific PCR primers for 86 repeats from five medically relevant microbial species. These PCR primers were tested experimentally. We demonstrate that using species-specific repeats as a PCR template region can increase the sensitivity of PCR in diagnostic tests. AVAILABILITY AND IMPLEMENTATION A web version of the method called MultiMPrimer3 was implemented and is freely available at (http://bioinfo.ut.ee/multimprimer3/).

DNA Fingerprinting Based on Repetitive Sequences of Iranian Indigenous Lactobacilli Species by (GTG)5- REP-PCR

Background and Objective: The use of lactobacilli as probiotics requires the application of accurate and reliable methods for the detection and identification of bacteria at the strain level. Repetitive sequence-based polymerase chain reaction (rep-PCR), a DNA fingerprinting technique, has been successfully used as a powerful molecular typing method to determine taxonomic and phylogenetic relat...

Detection and Discrimination of Theileria annulata and Theileria lestoquardi by using a Single PCR

The aim of this study was to detect and differentiate Theileria annulata and T. lestoquardi (hirci) by PCR. Members of the genus Theileria are tick-borne hemoprotozoan parasites those cause fatal and enervating diseases of cattle and sheep in Iran . In order to develop a specific method for detecting and identification of Theileria species, specific primers from the surface protein (SP) seque...

Specific Primers For Identification and Detection of Three Major Desert Truffle Species in Iran with Special Reference to Terfezia claveryi

ABSTRACT- Desert truffles are hypogeous ascomyceteous ectomycorrhizal fungi, occurring in arid and semi-arid ecosystems. A PCR-based method was developed for the identification of 3 major desert truffles of Iran: Terfezia claveryi, Tirmania pinoyi and Tirmania nivea based on internal transcribed spacers of rDNA. Two specific PCR primers were designed for T. claveryi, 4 for T. pinoyi, and 2 for ...

Inventory of new microbial taxa from Iran

Microorganisms constitute more than half of the Earth's biomass and in addition to quantity, they represent great diversity. In January 2013, the number of officially registered names forprokaryotic microorganisms at the genus and species levels were, respectively, approximately2100 and 12000. Current authorized methods for classification and identification of newmicrobial taxa are polyphasic ap...

Isolation and identification of Eurotium species from contaminated rice by morphology and DNA sequencing

30 milled rice samples were collected from retailers in four states of Malaysia. These samples were evaluated for Eurotium spp. contaminations by direct plating on malt extract salt agar (MESA). All Eurotium were isolated and identified based on morphology and nucleotide sequences of internal transcribed spacer 1 (ITS1) and ITS2 of the rDNA.  Four Eurotium species (E. rubrum, E. amstelodami, E....
